Taq DNA Polymerase

Gene Taqは、Thermus aquaticus YT1のDNAポリメラーゼ遺伝子をクローニングし、改変したものを大腸菌にて発現させ、分離精製した耐熱性DNAポリメラーゼであり、PCR法に適している。
Taq DNA ポリメラーゼのN末端を一部欠失させてあるため、内因性の5'→3'エキソヌクレアーゼ活性を有していない。また、特に1kbp以下のDNA断片の増幅効率が高い。
本品には、PCR法を行うために必要な基質dNTP混合液と10倍濃度の反応バッファー(10×Gene Taq Universal Buffer)が添付されている。

●活性	5 units/µl
●単位の定義	1 unitは、activated calf thymus DNAをプライマー/鋳型として74°C、30分間に10nmoleのデオキシヌクレオチドを酸不溶性沈殿物に取り込む酵素活性とする。
●形状	20mmol/l Tris-HCl(pH8.0), 100mmol/l KCl, 0.1mmol/l EDTA, 0.5% Tween20, 1mmol/l DTT, 50% Glycerol
●純度	本酵素10unitsと1µgのλ/Hind III digestとを74°C、1時間反応させてもアガロースゲル電気泳動パターンに変化は認められない。
●添付バッファー	[dNTP Mixture]	250unitsごとに1本(800µl)添付 (2.5mmol/l dATP, 2.5mmol/l dTTP, 2.5mmol/l dGTP, 2.5mmol/l dCTP)
	[10×Gene Taq Universal Buffer]	250unitsごとに1本(1ml)添付 (Mg2+濃度 15mmol/l)
