Taq DNA Polymerase

Gene Taq （ジーンタック）は、Thermus aquaticus YT1のDNAポリメラーゼ遺伝子をクローニングし、改変したものを大腸菌にて発現させ、分離精製した耐熱性DNAポリメラーゼであり、PCR法に適している。またターミナルトランスフェラーゼ活性を有しており、得られたPCR産物はTAクローニングに使用することができる。
Taq DNA ポリメラーゼのN末端を一部欠失させてあるため、5'→3'エキソヌクレアーゼ活性を有していない。また、特に1kbp以下のDNA断片の増幅効率が高い。

＜単位の定義＞	1 unitは、activated calf thymus DNAをプライマー/鋳型として74°C、30分間に10nmoleのデオキシヌクレオチドを酸不溶性沈殿物に取り込む酵素活性とする。
＜形状＞	20mmol/l Tris-HCl(pH8.0), 100mmol/l KCl, 0.1mmol/l EDTA, 0.5% Tween20, 1mmol/l DTT, 50% Glycerol
＜純度＞	本酵素10unitsと1µgのλ/Hind III digestとを74°C、1時間反応させてもアガロースゲル電気泳動のパターンに変化は認められない。
