ホットスタート DNA Polymerase

THUNDER Taq DNA Polymerase の活性領域を化学修飾することで酵素活性をコントロールしており、PCRサイクルに入る前に95℃、5～10分間の熱処理によって酵素の活性化処理を行います。ホットスタートとタイムリリースによって酵素を目的としたターゲットの増幅のみに集中させ、特異的で高品質なPCR産物の獲得に導きます。

【使用例①： 1.5kbp、3.0kbp、6.0kbpのフラグメントを増幅】

A社 ホットスタートTaq DNA Polymerase、THUNDER Taq Gold DNA Polymerase、THUNDER Taq DNA Polymerase、N社 Taq DNA Polymeraseをそれぞれ2.5Uを使用し、λDNAを鋳型として1.5kbp、3.0kbp 及び 6.0kbpを増幅した。

サイクル条件 (ABI 2720) 95℃ 10min.     95℃ 30sec. 25cycles 68℃ 6min. 68℃ 5min.     4℃

【使用例②：約500bpのフラグメントを増幅】

THUNDER Taq Gold DNA Polymeraseと他社のHot-Start PCR用酵素（A社、B 社、C 社）を使用し、特異性の低い3対のプライマー(a,b,c) を用いて約500bpのフラグメントを増幅した。

サイクル条件 (ABI 2720) 95℃ 10min.     95℃ 10sec. 30cycles 58℃ 30sec. 72℃ 1min. 72℃ 3min.     4℃
